How-To-Unlock:
To Unlock Ultra Instinct; When running default transformations, during the run, when you reach a Total Mastery Level of 80, a message will appear stating the hidden power is unlocked. On Singleplayer; when an attack could possibly kill you, you will instead trigger the hidden transformation. On Multiplayer, the transformation will trigger immediately. If you have additional transformations that have a Total Mastery Level requirement above 80, Ultra instinct will unlock at (Highest Transformation's Mastery Level + 10).
