In my company we don't use default port for sql server but the custom one. For example when I connect to named instance with listener I don't have to specify custom port number - just connect ListenerName\InstanceName. We do this with all our servers (around 100). SQL Browser is running everywhere.
Issue is with one of the new listener. When I connect to that one I have to type port number: ListenerName,12345\InstanceName - and I have no idea why it is like that just for this one. I checked all configuration and it is the same as other servers.

In general, when the port is the default port of 1433, then you do not have to specify a port number when connecting to your listener. If you specify a custom port, the client must explicitly specify the port.
The AG listener port is different from SQL Server instance port
